有機(jī)溶劑萃取法或浸泡提取法是提取大豆異黃酮的一種常用的提取方法。本實驗以乙醇為溶劑,大豆豆渣為原料,研究了大豆異黃酮的提取工藝,選取了濃度、提取時間、液料比、溫度四個變量對大豆異黃酮提取工藝的影響,并進(jìn)行了響應(yīng)曲面法優(yōu)化分析,建立了相關(guān)數(shù)學(xué)模型。研究結(jié)果表明,乙醇濃度、溫度、提取時間、液料比對豆渣中大豆異黃酮的提取均產(chǎn)生不同程度的影響。從實驗結(jié)果得知,乙醇濃度對豆渣異黃酮提取量的影響最大,提取時間對大豆異黃酮提取量的影響最小。通過響應(yīng)曲面法得出大豆異黃酮提取的最佳工藝條件,乙醇濃度70%、溫度60℃、提取時間5h、液料比16mL/g。此時提取量可達(dá)7.72mg/10g,與實驗值7.68mg/10g基本吻合。因此,該實驗方案具有一定的可行性,該模型具有一定的理論價值,可為相關(guān)工業(yè)生產(chǎn)提供一定的理論依據(jù)。

Optimization of isoflavones extraction from soybean dregs using response surface methodology
ABSTRACT
Soybean isoflavones from soybean extracted trace active substances,has many important physiological functions.Bean dregs as experimental material,this paper studied the soybean isoflavone extraction technology.
Organic solvent extraction or soaking extraction is a king of commonly used to extract soybean isoflavone extraction method.This experiment with ethanol as solvent,soy bean dregs as raw material,soybean isoflavone extraction technology was studied and selected the concentration,extraction time and solid-liquid ratio,temperature four variables impact on soybean isoflavone extraction technology,and has carried on the response surface method optimization analysis,the relevant mathematical model is established.The research results show that ethanol concentration,temperature,extraction time and solid-liquid ratio on bean dregs in the extraction of soybean isoflavones are varying degress of impact.From the experiment results indicate that the ethanol concentration affects the quantity of bean dregs isoflavone extraction,the largest amount of time for extracting soybean isoflavines minimal impact. By response surface method for optimum process condition of soybean isoflavones extract,ethanod concentration 70%,5h temperature 60℃, extraction time, liquid than 16 ml/g. Extract the quantity at this time can be up to 7.72 mg/g, with the experimental value of 7.68 mg / 10 g. This experiment scheme has certain feasibility, therefore, has certain theory value, the model can provide certain theoretical basis for related industrial production.
