水華魚腥藻乙酰乳酸合成酶的提取及其活性的測(cè)定

摘 要
本實(shí)驗(yàn)利用超聲波細(xì)胞破碎提取了水華魚腥藻中的乙酰乳酸合成酶（ALS），并且通過與乙酰甲基甲醇（OD525）標(biāo)準(zhǔn)曲線的對(duì)照測(cè)定了乙酰乳酸合成酶的比活力，其值為168.94 (A525nm nmol/mg(pro.)*h)。本實(shí)驗(yàn)利用不同濃度的丙酮酸鈉酶反應(yīng)液測(cè)定了不同底物濃度下乙酰乳酸合成酶的比活力，得出乙酰乳酸合成酶的米氏常數(shù)Km＝8.17mmol/L。

ABSTRACT
This experiment ueses ultrusonic cell disrupter system to pick up acetolactate synthases in anabaena flos-aquaes.According to the comparison between the standard data and measured data, we get the specific activity of anabaena flos-aquae at 168.94 (A525nm nmol/mg(pro.)*h).Our experiment also makes use of the solutions with the different concentrations of sodium pyruvate to mensurate the specific activity in each concentration.The michaelis constant of the acetolactate synthase we get through this experiment is 8.17mmol/L.
