摘 要:采用ISSR技術(shù)對廣東省深圳市周圍8個(gè)薇甘菊自然種群的遺傳多樣性進(jìn)行分析。3個(gè)引物共檢測到37個(gè)位點(diǎn),其中多態(tài)位點(diǎn)30個(gè),多態(tài)位點(diǎn)百分率81.08%。物種水平的Shannon信息指數(shù)為0.4172,Nei指數(shù)為0.2808,表明物種水平的遺傳多樣性很高;種群水平的遺傳多樣性也較高,多態(tài)位點(diǎn)百分率平均為57.43%,Shannon信息指數(shù)平均為0.3613,Nei指數(shù)平均為0.2521。AMOVA分子變異顯示4.41% 的變異存在于種群間,95.59%的變異存在于種群內(nèi)。種群間的基因流較高,為4.3946,遺傳相似度平均為0.9564,遺傳距離平均為0.0448。聚類分析顯示,8個(gè)薇甘菊自然種群的遺傳距離與地理距離存在一定的相關(guān)性。

Genetic Diversity in the Populations of Mikania micrantha Revealed by ISSR Molecular Markers
Abstract: Genetic diversity in the eight populations of Mikania micrantha around Shenzhen city, Guangdong province were analyzed using inter simple sequence repeat (ISSR) molecular markers.A total of 37 loci, 30 of which were polymorphic loci, the percent of polymorphic loci was 81.08% in the species, while Shannon,s information index and Nei,s gene diversity were 0.4172 and 0.2808, respectively. These indicated that the genetic diversity of Mikania micrantha was relatively high. The mean value of the percent of polymorphic loci, Shannon,s information index and Nei,s gene diversity were 57.43%, 0.3613 and 0.2521, respectively. Analysis of molecular variance (AMOVA) demonstrated that the among-population component accounted for 4.41% of the total variations, while the within-population component accounts for 95.59%. The gene flow was high (4.3946), the genetic similarity and genetic distance with an average of 0.9564 and 0.0448, respectively. The clustering analysis showed that there is a certain correlation between genetic distance and geographic distance of eight natural populations of Mikania micrantha .
